Antigen-specific CD4+ TH1 and CD8+ CTL cells play a significant role in intracellular infection and the occurrence and development of tumors, thus becoming an important part of immunological research and vaccine development for related diseases. Protein antigens (including inactivated pathogenic microorganisms, tumor cell lysates, recombinant or extracted protein antigens, synthetic peptides, etc.) cannot effectively induce antigen-specific CD4+ TH1 and CD8+ CTL responses under the action of conventional adjuvants such as aluminum hydroxide. Quick CTL cellular immune adjuvant is a new type of water-soluble cellular immune adjuvant with independent intellectual property rights and a unique formula. It can be used as a protein antigen adjuvant to immunize mice and induce effective antigen-specific CD4+ TH1 and CD8+ CTL responses.
two Usage method
Dilute the antigen to twice the final concentration with normal saline (prepare at a dosage of 50μl per injection). Note: The recommended dosage of antigen used is (1) for synthetic peptides with relatively weak immunogenicity, 10-100μg per dose; (2) Subunit protein antigen: 1-10μg per injection; (3) Inactivated pathogenic microorganisms and virus-like particle antigens with strong immunogenicity: 1-10μg per injection. (4) Tumor cell lysate: 10-100μg per injection.
2. Thaw the adjuvant in a 37 ° C water bath, mix thoroughly, and under aseptic conditions, take out the required amount (50μl per injection) and mix it with the antigen in a volume ratio of 1:1.
3. Inject 100μl of immunized mice subcutaneously through the calf muscle of the hind leg or the groin.
On the 7th day, give a booster shot in the same way. Note: The adjuvant and antigen should be prepared and used immediately each time
5. On the 14th day, antigen-specific CD4+ TH1 and/or CD8+ CTL responses in the spleen or draining lymph nodes were detected by ELISPOT, intracellular cytokine staining, MHC tetramer staining or other feasible methods.
Precautions
This product is designed for immunizing mice, but in theory, it can also be used on some other experimental animals. If the customer intends to use it for other experimental animals, they should determine its usability and the dosage of adjuvant through pre-experiments by themselves.
2. This product is a potent cellular immune adjuvant. If no specific cellular immune response is detected after immunization, please pay close attention to the quality of the antigen and the detection method.
3. This product is for animal experimentation only and must not be used for human immunization.
