Product Introduction

The SDS-PAGE Color Rapid Gel Preparation Kit (10%) adopts a formula in the form of a premix. When preparing the gel, only the improved coagulant needs to be added, which is convenient and fast. The upper layer of gel is a colored (purple) gel, which is convenient for spotting. The gel prepared by this product can also be used in non-denatured PAGE gel electrophoresis experiments.
This kit is equipped with an improved coagulant accelerator, which has no pungent smell of TEMED, good stability and excellent catalytic effect. It only takes about twenty minutes to prepare one or more gels. For ease of use, the improved coagulant that has been opened can be stored at 4℃ for at least 3 months.

Operation steps
 
1. According to the thickness of the gel to be prepared, take equal volumes of the lower gel solution and the lower gel buffer solution, and mix them evenly. Add the corresponding volume of the improved coagulant accelerator and mix well.
2. Immediately pour the prepared lower layer of glue into the glass plate so that the distance between the liquid surface and the upper edge of the short glass plate is 0.5 cm longer than the comb teeth. Add an appropriate amount of water or anhydrous ethanol to seal the liquid. Wait for the lower layer of glue to solidify for about 10 to 15 minutes (when a clear boundary line can be seen between the liquid seal solution and the glue, it indicates that the gelatin has solidified), and then pour out the upper layer solution.
Note: 1) The volume of the solution prepared is in excess. A small amount can be left in the tube for easy observation of the gel state.
2) When the room temperature is relatively low, the solidification speed is slower, and the gel time needs to be appropriately extended.
3. According to the thickness of the gel to be prepared, take equal volumes of the upper layer gel solution and the colored upper layer gel buffer solution, and mix them evenly. Add the corresponding volume of the improved coagulant accelerator and mix well.
4. Immediately pour the prepared upper layer glue into the glass plate, insert the comb, and wait for the upper layer glue to solidify (5-10 minutes) before removing the comb.

Note: 1) Try to use freshly prepared electrophoresis buffer solution;
2) Recommended electrophoresis conditions: 150V, approximately 50 minutes or 200 V, approximately 35 minutes.

Precautions
 
1. The upper gel prepared by this product has no concentration effect on the sample and is similar to pre-prepared gel. Compared with traditional PAGE gel, it has a better separation effect on egg white bands. Small molecule proteins (such as 10 kDa) can also be clearly separated, and the protein bands are narrower and sharper.
2. The dosage of the improved coagulant can be adjusted according to personal habits and experience. Adding more coagulant can accelerate gelation, and vice versa. Under the same conditions, the higher the temperature, the faster the gelation speed. When the temperature is too high, it is recommended to appropriately reduce the dosage of the accelerator.
3. Restoring the pre-gel solution and buffer to room temperature can effectively prevent the formation of bubbles during the gelation process.
4. For your safety and health, please wear a lab coat and disposable gloves when operating.
